TPI 287 is a new anti-microbubule agent manufactured from naturally occurring taxanes, but modified to overcome resistance to taxanes and bind to mutant tubulin. In laboratory experiments it was greatly more active against multidrug-resistant cell lines than other compounds tested, and more active in animal model tumors of multidrug-resistant cell lines. Phase I testing showed little or no toxicity from TPI 287, which is able to cross the blood-brain barrier and achieve pharmacologically relevant levels. |
