NO prior gene therapy, viral based therapy
NO tumor involvement which would require ventricular, brainstem, basal ganglia or posterior fossa inoculation
IF on steroids, must be on stable regimen for 2 weeks prior to injection
NO active infection (HIV, hepatitis, herpes)
Comments: G207, a Genetically Engineered Herpes simplex virus, type 1, being used as a viral oncolytic therapy in the treatment of malignant glioblastoma. The virus has been neuroattenuated by the
removal of two copies of the g34.5 gene and a disabling intersertion of the lacZ marker gene in the ICP 6 region. It remains sensitive to ganciclovir and other anti-herpetic agents, as well as it
temperature sensitive.

A Phase 2 Study To Evaluate The Efficacy And Safety Of Peretitive Daily Intravenous Doses Of RSR13 Administered To Patients Receiving Cranial Radiation Therapy For Glioblastoma Multiforme.

Comments: RSR13 is being studied for its potential use as an enhancement to standard brain radiation therapy for glioblastoma multiforme. RSR13 causes a change in the way that oxygen binds to blood.
This change results in more oxygen being released into the body tissues, including brain tissues. Tumor tissues can have areas of low oxygen concentrations, which may cause radiation therapy to be
less effective. By increasing the oxygen in tumor tissue, the effectiveness of radiation treatment on the tumor may be improved. Patients must have a histologically confirmed GBM, radiation therapy
must begin within six weeks of GBM diagnosis, must have adequate pulmonary function and laboratory tests within specified ranges, must have no other serious medical ilness or current malignancy.
Patients who are eligible and enrolled in the study will have a central access device placed and receive an intravenous infusion of RSR13 over a 30-minute period each day just prior to
receiving radiation therapy. Thirty treatments of RSR13 and radiation therapy will be given over a six-week period. Patients will be evaluated for treatment effectivenss with brain scans and
physical examinations every one-two months after the treatment period. This study is being conducted in conjunction with the New Approaches

Phase 3 Study Of Crisnatol Mesylate Vs. BCNU In Patients With Glioblastoma Multiforme

Comments: Crisnatol has been shown to have activity in patients with glioblastoma
multiforme. This study is a multinational, randomized, phase 3 trial of crisnatol mesylate versus BCNU in the consolidative treatment of patients with glioblastoma multiforme. Patients will be
randomized to receive either BCNU (IV infusion, every 6 weeks) or crisnatol (continuous IV infusion for 72 hours, every 3 weeks). Both treatments may be given on an outpatient basis. Patients must
begin treatment for at least 2 weeks, but no more than 4 weeks after completing radiation therapy. Treatment may
continue up to one year or until disease progression.
Patients 18 years of age and older with histologically confirmed diagnosis
of glioblastoma multiforme (grade IV astrocytoma) who have received at least 5500-6000 cGy of radiation therapy to the primary tumor, with or
without surgical resection (resection recommended when clinically possible)
No prior chemotherapy for their brain tumor
SWOG performance status of 0-2
Pathology report/slides and radiation therapy summary/films must be reviewed
by designated consultants to determine eligibility

No Brainstem Location.
Comments: OKT-3 is a synthetic serum antibody. It is an FDA approved medication, made by state-of-the-art laboratory methods. This antibody reacts with a special protein on the surface of a certain
white blood cell - the T-lymphocytes. It has been used in the past, at high dose, to counter the organ rejection in patients with kidney or other organ transplants. In this study, it is used at much
lower doses, which has the opposite effect - it is an immune system stimulator.
It increases the number of t-lymphocytes and alters their behavior to increase their cancer fighting ability.
It is given by IV infusion over 60 minutes. 24 hours later, Cyclophosphamide is used, which also stimulates the immune system. This is done in a 2 - 3 day hospital stay. Treatments are repeated at
4 week intervals.

Comments: Hypericen is a naturally occurring derivative of the plant rauwolfia which is being synthesized for medical uses in the United States by VimRx Corporation. Hypericen has been shown in
cultures and animal models to have antiglioma activity.
USC is recruiting patients for a study of the efficacy of hypericen in the treatment of malignant gliomas. The medication is administered orally for 3 months. Patients are asked to come in daily for
the first month to check for symptoms of photosensitivity (tingling and itching of the skin on exposure to sunlight) which is the only so far described toxicity of hypericen. Serial MR scanning to
access for tumor progression is part of the protocol.

Comments: RSR13 is being studied for its potential use as an enhancement to standard brain radiation therapy for glioblastoma multiforme. RSR13 causes a change in the way that oxygen binds to
blood. This change results in more oxygen being released into the body tissues, including brain tissues. Tumor tissues can have areas of low oxygen concentrations, which may cause radiation therapy
to be less effective. By increasing the oxygen in tumor tissue, the effectiveness of radiation treatment on the tumor may be improved. Patients must have a histologically confirmed GBM, radiation
therapy must begin within six weeks of GBM diagnosis, must have adequate pulmonary function and laboratory tests within specified ranges, must have no other serious medical ilness or current
malignancy. Patients who are eligible and enrolled in the study will have a central access device placed and receive an intravenous infusion of RSR13 over a 30-minute period each day just prior to
receiving radiation therapy. Thirty treatments of RSR13 and radiation therapy will be given over a six-week period. Patients will be evaluated for treatment effectivenss with brain scans and
physical examinations every one-two months after the treatment period. This study is being conducted in conjunction with the New Approaches to Brain Tumor Therapy.

Comments: Crisnatol has been shown to have activity in patients with glioblastoma multiforme. This study is a multinational, randomized, phase 3 trial of
crisnatol mesylate versus BCNU in the consolidative treatment of patients
with glioblastoma multiforme. Patients will be randomized to receive either BCNU (IV infusion, every 6 weeks) or crisnatol (continuous IV
infusion for 72 hours, every 3 weeks). Both treatments may be given on an outpatient basis. Patients must begin treatment for at least 2 weeks, but no more than 4 weeks after completing radiation
therapy. Treatment may continue up to one year or until disease progression.
Patients 18 years of age and older with histologically confirmed diagnosis
of glioblastoma multiforme (grade IV astrocytoma) who have received at least 5500-6000 cGy of radiation therapy to the primary tumor, with or
without surgical resection (resection recommended when clinically possible)
No prior chemotherapy for their brain tumor
SWOG performance status of 0-2
Pathology report/slides and radiation therapy summary/films must be reviewed
by designated consultants to determine eligibility

Phase 3 Study Of Crisnatol Mesylate Vs. BCNU In Patients With Glioblastoma Multiforme

Comments: Crisnatol has been shown to have activity in patients with glioblastoma multiforme. This study is a multinational, randomized, phase 3 trial of
crisnatol mesylate versus BCNU in the consolidative treatment of patients with glioblastoma multiforme. Patients will be randomized to receive either BCNU (IV infusion, every 6 weeks) or crisnatol
(continuous IV
infusion for 72 hours, every 3 weeks). Both treatments may be given on an outpatient basis. Patients must begin treatment for at least 2 weeks, but no more than 4 weeks after completing radiation
therapy. Treatment may continue up to one year or until disease progression.
Patients 18 years of age and older with histologically confirmed diagnosis of glioblastoma multiforme (grade IV astrocytoma) who have received at least 5500-6000 cGy of radiation therapy to the
primary tumor, with or without surgical resection (resection recommended when clinically possible)
No prior chemotherapy for their brain tumor
SWOG performance status of 0-2
Pathology report/slides and radiation therapy summary/films must be reviewed
by designated consultants to determine eligibility
